The 26-year-old returned from a post-Confederations Cup holiday to join Jose Mourinho's squad for their tour to the United States and played 30 minutes of the match with Inter Milan last Thursday.
However, Luiz did not train with the rest of the squad today and has been ruled out of the Real match with a suspected hamstring injury.
Chelsea wrote on Twitter: 'While the other lads trained, @DavidLuiz-4 completed his own workout. He won't play against Real tomorrow.'
The former Benfica star signed for the West London club for £21million in 2011 and is a target for Barcelona this summer.
The Spanish giant's director of football Andoni Zubizaretta has confirmed a centre-back is their top priority although they do have other options if Chelsea continue to price Luiz out of a deal, valuing the player at £45m.
